Studies of alloantibody effect on skin graft survival in mice.

چکیده
Alloantisera (anti-donor and anti-recipient alloantiserum) were prepared by skin grafting followed four inoculations of 1 x 107 splenocytes. These alloantisera were cytotoxic ( x 64) to target cells specifically. Anti-recipient alloantiserum was effective on enhancing skin graft survival in mice as same as anti-donor alloantiserum, when administered on the operative day, second postoperative day and fourth postoperative day between 50-200 μl at one time. Diversity of gut microflora is required for the generation of B cell with regulatory properties in a skin graft model
B cells have been reported to promote graft rejection through alloantibody production. However, there is growing evidence that B cells can contribute to the maintenance of tolerance. Here, we used a mouse model of MHC-class I mismatched skin transplantation to investigate the contribution of B cells to graft survival. We demonstrate that adoptive transfer of B cells prolongs skin graft survival...
متن کاملEffect of Mesenchymal Stem Cells on ILT3 Expression in the Splenocytes of Skin Graft Recipient Mice
Background: Mesenchymal stem cells (MSCs) are considered as effective therapeutic cells in transplantation due to their immunomodulatory activities. However, precise mechanism of MSCs immunomodulatory activity is not completely understood. Objectives: To study the role of Immunoglobulin-like transcripts-3 (ILT3) immunomodulatory receptor in immune tolerance induced by MSCs in skin transpla...
متن کاملStudies investigating pretransplant donor-specific blood transfusion, rapamycin, and the CD154-specific antibody IDEC-131 in a nonhuman primate model of skin allotransplantation.
Anti-CD154 variably prolongs allograft survival in nonhuman primates. Rodent studies suggest that adding pretransplant donor-specific transfusion (DST) and/or rapamycin to anti-CD154 improves survival. The CD154-specific Ab IDEC-131 was tested alone and in combination with rapamycin for its ability to inhibit rhesus MLRs. The ability of the Ab to block endothelial activation was also assessed. ...
متن کاملEffect of alloantibodies on corneal allograft survival.
PURPOSE The precise role of antibodies in corneal transplantation is ambiguous, with evidence to support as well as repudiate their involvement in graft rejection. Accordingly, this study was undertaken to investigate the direct contribution of donor-specific antibodies to corneal graft rejection. METHODS Serum samples from CB6F1 rejecters of orthotopically grafted C3H/Hej corneas were tested...
متن کاملCD4+ T cell-mediated rejection of major histocompatibility complex class I-disparate grafts: a role for alloantibody.
Experimental studies of the T cell requirement for rejection of class I major histocompatibility complex (MHC)-disparate grafts have generated controversy over both the autonomy of CD8+ T cells and the mechanism whereby CD4+ T cells are able to independently mediate rejection. In this study of rejection of RT1Aa class I MHC-disparate rat cardiac and skin allografts by high-responder PVG RT1u re...
